Here is how I would do it
1. Sign ups complete
2. Two Players will be chosen (if this is a game for 24 players, 26 sign up, the extra two become these two) chose teams. These players do not participate in the game, but mentor everyone on their team.

I have given advice to players after they have died in my games (e.g. Artifice 101), but it makes sense to clarify any confusing terms and/or abbreviations to players as well. However, if there is at least one experienced player in the game, that player should be able to guide the newer players along...though that player could be manipulative scum!

Mafiascum, yes. Whenever I play in Basics, I tend to follow that code of ethics. I'm always playing to win, but in a basic, I'm also there to help the new players. Most of the time I'd answer their questions honestly, and if for whatever reason I had to lie, I would explain immediately after the game ended.

The thing about this is if that player hits l-2 later on in the game and claims doctor they will likely be lynched for lying.

Because there's plenty of mods that don't include doctor. Look at Ged's game for the perfect example - a scum claimed doc and rode that wave to the end of the game.
Seppel also has modded two games and neither one included a doc. And, in Indiana Jones, the scum did have a doc and rode that claim ALMOST to the end. He ended up accidentally killing himself, though. Most awesome game ending ever.

Actually, looking again, neither of mine nor my husband's basics had docs in them, either. I had a bodyguard in Angel and a jailer in Twilight, and he had a bodyguard in Snow Valley and Super Sentai.

While i have absolutlely no idea on RL mafia games, id like to ask to review. My only justifiable reason is that since i have no idea, ill be in the same position as a lot of you pratcipants.

Make 8 cards and explain them:
2x [MAF] (MAFIA: At night, the Mafia wakes up, identify each other, and collectively choose someone to kill.)
1x [DOC] (DOCTOR: At night, chooses someone to protect. If they protect the person who the mafia kill, that person doesn't die. Cannot self-protect.)
1x [COP] (COP: At night, the Cop wakes up and chooses someone to investigate. Thumbs up from the moderator means "good guy," thumbs down means "bad guy.")
3x [VIL] (VILLAGER: At night, does nothing.)
1x [M0D] (MODERATOR: Moderates the game. You keep this for the first few games, and then let someone else moderate if they want.)
Shuffle and hand out a card to everyone. Then everyone places the card face down in front of them. They are forbidden from touching their card for the rest of the game.
NIGHT
At night, everyone should close their eyes and smack one hand on their knee to create enough noise to drown out the quiet (but audible) sounds of people moving. To make things easier, it's best if you start with a Night 0 where players can kill, protect, and investigate.
The moderator says:
[1] "Mafia, wake up, identify each other, and choose someone to kill." [Mafia points at someone. REMEMBER WHO THAT PERSON IS, AND REMEMBER WHO THE MAFIA ARE.] "Mafia go to sleep."
[2] "Doc, wake up and choose someone to protect." [Doc points at someone. If it's the same person the Mafia chose, that person won't die.] "Doc go to sleep."
[3] "Cop, wake up and choose someone to investigate." [Cop points at someone. You give the cop a thumbs up or thumbs down. Don't screw it up!] "Cop go to sleep."
DAY
"Everyone wake up!"
[If someone is dead...] "Except for you." [...reveal dead person's card.] "[Name] was a [Role]! Discuss and find the murderers amongst you!"
Players may now talk and accuse each other.
There are multiple ways to accuse, and it can be done based on your moderating preference:
[1] One person raises one hand and points at someone, signaling an accusation. If another person "seconds" that vote, the accused player is put up on the stand to defend himself or herself. Then players may discuss, ask questions, and then a vote is called. A majority vote causes that person to be lynched. No majority returns the game to the accusation stage. If nobody is lynched after three defenses, the day ends with a no-lynch.
[2] The town gets a reasonable amount of time to discuss everything, then a vote is called. You go around the circle and ask who wants to vote [name], then tally up the votes. The two players with the most votes defend themselves, then a second vote is called. The player with the most votes of those two is lynched.
[3 (My preferred method; works best for crowds of 12 or less)] Players raise their hands and point (like in method [1]), but instead of accusing, a finger pointing means a vote. The instant someone reaches a majority of votes, that player is lynched.
As soon as a lynch has been made, everyone must stop talking and immediately go to sleep. This almost never happens with a social group, so you'll have to say, "Go to sleep!" about 3 times.
Then it is night again.
The town wins when the mafia are dead.
The mafia wins when the mafia equal or outnumber the town.
Tell people that players who have died still win the game. Lots of people think they lose when they die, so it's important to stress that this is a team game.
AND THAT'S ALL THERE IS TO REAL LIFE MAFIA. PREPARE YOURSELF FOR 5 OR 6 GAMES OF THE TOWN LOSING TERRIBLY BEFORE THEY START GETTING GOOD ENOUGH TO CATCH MAFIA. OF THE 12 GAMES OF MAFIA I'VE HOSTED IN THE PAST MONTH, THE TOWN HAS WON ONLY TWICE. PROTIP: THE WOMEN IN MY GROUP ARE VERY GOOD AT BEING MAFIA.
If players get more experienced or you have more people playing, add two new roles:
1x [VIG] (Vigilante. At night, can kill someone. Advise the vig that it is probably best if he does not kill on the first night. Call vig before doctor.)
1x [RB] (Roleblocker. At night, can stop someone's power from working for that night. After the mafia choose who to kill, ask who the roleblocker would like to block. Allow the roleblocker to block and kill in the same night if he's the only Mafia left alive.)
Gandening_dan may know something about that too. Also i suggest you buy "ultimate werewolf" not for the cards but mainly for the manual that has some very good tips.

The game is not being dumbed down. Control is doing fine; Draw-Go is not the only kind of control. Aggro is doing fine; Red Deck Wins is not the only kind of aggro. Creature combat is an important core concept and belongs in every color. Mythic rarity is not destroying the game. People whine too much for no good reason. Magic is more popular than ever, so keep calm, brew some decks and play some damn cards.
Yea, I can be of help here. To keep in flavor, we used Magic cards, since 90% of players in our group play MTG. I go about moderating almost the exact same way as Seppel described.
In our larger games, Mafia had a RB, rolecop & godfather. I used a thumb across the throat symbol to kill, a monacle for the rolecop, and wings (our RB is a bat) for the RB. I found it least revealing to say nothing during the mafia phase besides "Mafia, wake up" and "Go to sleep," taking about the same amount of time to not let the town know which roles were dead. Choosing to let mafia use more than one ability a night is up to you, I've done both (after PR's are dead) to some level of success.
If anyone opens their eyes at night, modkill them on the spot. If anyone sees another person's card, emphasize that they need to say so immediately. Make sure you call every role at night if you choose not to reveal roles on death (both versions work effectively, IMO). Having paper/pen or a laptop can make more complex games easier, as well as keeping track of who targeted who for endgame purposes, since people do get curious.
I've designed games with an SK, doublevoter (this can be very sneaky), survivor, vig, cult (did not go well), JoAT, jester, redirector (WAY too strong), and masons/lovers, so ship the setup if you feel so inclined.
Separate groups of friends around the circle to maximize interaction. Fingerpointing=vote. If a lynch isn't decided upon when time runs out, everyone has to vote without speaking. I've played that the town has to actively decide "we want to no-lynch" or something to that extent. The town sucks, generally, and like Seppel said, women are at a huge advantage.

I went to this after party for a play with my girlfriend at the time. We were all pretty drunk. Basically what happened was someone took a break-away glass object and smashed it over someone's head.

I've noticed nothing in particular either way. You need talkative people to make the game work, though, as well as being pretty sociable. Thespians also really love it.
Most conflict has come when there's something obvious to the group that someone doesn't catch on to, and it's never been more than a brief shouting match.

Done this before, as well as with my eyes closed, as well as while listening to music at high levels (eliminating senses) (not all at the same time). No real difference noted. I'm not good at finding "tells" so to speak.

I figure the, "So who's list did he choose?" method isn't too breakable, especially if chosen randomly. I do not envy the WIFOM the scum Captain would have to avoid after about 2 nights if they were a well respected/feared player in the subforum. Then again, scum could use that to lynch the virtually unlynchable....

They are not allowed to give reads or recommendations, but general advice to the players. I doubt it would hurt to encourage players to actively engage the mod who's still free to tell them they can't comment about certain things. I think it would be a healthy habit to newer players who (flatly) get intimidated about bothering the mod sometimes.
It'd be extra work for the mod, but modding a basic is piss easy.
I have given advice to players after they have died in my games (e.g. Artifice 101), but it makes sense to clarify any confusing terms and/or abbreviations to players as well. However, if there is at least one experienced player in the game, that player should be able to guide the newer players along...though that player could be manipulative scum!
The experienced players are limited to 2 in those games IIRC, and they have to meet some qualification. Oh yeah! They call them "IC's: Inexperience Challanged" players. Mafiascum? Maybe.
I think if a player is new to MTGS, mods of Basics should ask and direct them to at least the glossary and roles if not the Mafia Role Flash. Tends to help them get a foothold. We have a TON of jargon here.

Mafiascum, yes. Whenever I play in Basics, I tend to follow that code of ethics. I'm always playing to win, but in a basic, I'm also there to help the new players. Most of the time I'd answer their questions honestly, and if for whatever reason I had to lie, I would explain immediately after the game ended.

The thing about this is if that player hits l-2 later on in the game and claims doctor they will likely be lynched for lying.

Of course, just because it's screwball doesn't mean it's fun; anyone can come up with weird ideas for a game, but in the end what matters is coming up with fun ideas.
So please if anyone has any experience with Real Life Mafia and can help me review a very basic, and rather unorthodox setup i would be deeply grateful.

1x [DOC] (DOCTOR: At night, chooses someone to protect. If they protect the person who the mafia kill, that person doesn't die. Cannot self-protect.)
1x [COP] (COP: At night, the Cop wakes up and chooses someone to investigate. Thumbs up from the moderator means "good guy," thumbs down means "bad guy.")
3x [VIL] (VILLAGER: At night, does nothing.)
1x [M0D] (MODERATOR: Moderates the game. You keep this for the first few games, and then let someone else moderate if they want.)
Shuffle and hand out a card to everyone. Then everyone places the card face down in front of them. They are forbidden from touching their card for the rest of the game.
NIGHT
At night, everyone should close their eyes and smack one hand on their knee to create enough noise to drown out the quiet (but audible) sounds of people moving. To make things easier, it's best if you start with a Night 0 where players can kill, protect, and investigate.
The moderator says:
[1] "Mafia, wake up, identify each other, and choose someone to kill." [Mafia points at someone. REMEMBER WHO THAT PERSON IS, AND REMEMBER WHO THE MAFIA ARE.] "Mafia go to sleep."
[2] "Doc, wake up and choose someone to protect." [Doc points at someone. If it's the same person the Mafia chose, that person won't die.] "Doc go to sleep."
[3] "Cop, wake up and choose someone to investigate." [Cop points at someone. You give the cop a thumbs up or thumbs down. Don't screw it up!] "Cop go to sleep."
DAY
"Everyone wake up!"
[If someone is dead...] "Except for you." [...reveal dead person's card.] "[Name] was a [Role]! Discuss and find the murderers amongst you!"
Players may now talk and accuse each other.
There are multiple ways to accuse, and it can be done based on your moderating preference:
[1] One person raises one hand and points at someone, signaling an accusation. If another person "seconds" that vote, the accused player is put up on the stand to defend himself or herself. Then players may discuss, ask questions, and then a vote is called. A majority vote causes that person to be lynched. No majority returns the game to the accusation stage. If nobody is lynched after three defenses, the day ends with a no-lynch.
[2] The town gets a reasonable amount of time to discuss everything, then a vote is called. You go around the circle and ask who wants to vote [name], then tally up the votes. The two players with the most votes defend themselves, then a second vote is called. The player with the most votes of those two is lynched.
[3 (My preferred method; works best for crowds of 12 or less)] Players raise their hands and point (like in method [1]), but instead of accusing, a finger pointing means a vote. The instant someone reaches a majority of votes, that player is lynched.
As soon as a lynch has been made, everyone must stop talking and immediately go to sleep. This almost never happens with a social group, so you'll have to say, "Go to sleep!" about 3 times.
Then it is night again.
The town wins when the mafia are dead.
The mafia wins when the mafia equal or outnumber the town.
Tell people that players who have died still win the game. Lots of people think they lose when they die, so it's important to stress that this is a team game.
AND THAT'S ALL THERE IS TO REAL LIFE MAFIA. PREPARE YOURSELF FOR 5 OR 6 GAMES OF THE TOWN LOSING TERRIBLY BEFORE THEY START GETTING GOOD ENOUGH TO CATCH MAFIA. OF THE 12 GAMES OF MAFIA I'VE HOSTED IN THE PAST MONTH, THE TOWN HAS WON ONLY TWICE. PROTIP: THE WOMEN IN MY GROUP ARE VERY GOOD AT BEING MAFIA.
If players get more experienced or you have more people playing, add two new roles:
1x [VIG] (Vigilante. At night, can kill someone. Advise the vig that it is probably best if he does not kill on the first night. Call vig before doctor.)
1x [RB] (Roleblocker. At night, can stop someone's power from working for that night. After the mafia choose who to kill, ask who the roleblocker would like to block. Allow the roleblocker to block and kill in the same night if he's the only Mafia left alive.)

Mythic rarity is not destroying the game. People whine too much for no good reason. Magic is more popular than ever, so keep calm, brew some decks and play some damn cards.
In our larger games, Mafia had a RB, rolecop & godfather. I used a thumb across the throat symbol to kill, a monacle for the rolecop, and wings (our RB is a bat) for the RB. I found it least revealing to say nothing during the mafia phase besides "Mafia, wake up" and "Go to sleep," taking about the same amount of time to not let the town know which roles were dead. Choosing to let mafia use more than one ability a night is up to you, I've done both (after PR's are dead) to some level of success.
If anyone opens their eyes at night, modkill them on the spot. If anyone sees another person's card, emphasize that they need to say so immediately. Make sure you call every role at night if you choose not to reveal roles on death (both versions work effectively, IMO). Having paper/pen or a laptop can make more complex games easier, as well as keeping track of who targeted who for endgame purposes, since people do get curious.
I've designed games with an SK, doublevoter (this can be very sneaky), survivor, vig, cult (did not go well), JoAT, jester, redirector (WAY too strong), and masons/lovers, so ship the setup if you feel so inclined.
Separate groups of friends around the circle to maximize interaction. Fingerpointing=vote. If a lynch isn't decided upon when time runs out, everyone has to vote without speaking. I've played that the town has to actively decide "we want to no-lynch" or something to that extent. The town sucks, generally, and like Seppel said, women are at a huge advantage.

I went to this after party for a play with my girlfriend at the time. We were all pretty drunk. Basically what happened was someone took a break-away glass object and smashed it over someone's head.

Most conflict has come when there's something obvious to the group that someone doesn't catch on to, and it's never been more than a brief shouting match.
Note this is only one case. We have a staff party for the school I'm working at, and knowing some of the teachers, heavy amount of alcohol will be consumed, I'll let you know how that goes.
I've foudn it's fine in conjunction with other substances.

Done this before, as well as with my eyes closed, as well as while listening to music at high levels (eliminating senses) (not all at the same time). No real difference noted. I'm not good at finding "tells" so to speak.
